Measurement of N-Methylcarbamoyloximes and N-Methylcarbamates in Water by Direct Aqueous Injection HPLC with Postcolumn Derivatization


This method is for the determination of N-methylcarbamoyloximes and N-methylcarbamates in finished drinking waters.  The collected sample is filtered and an aliquot up to 100uL is injected into a high performance liquid chromatograph (HPLC) equipped with a reversed phase (C18) column.  After the analytes elute from the column, the analytes are hydrolyzed in a postcolumn reaction with 0.075 N sodium hydroxide (NaOH) at 80 to 100°C to form methyl amine.  Methyl amine is reacted with o-phthalaldehyde (OPA) and 2-mercaptoethanol (or N,N-dimethyl-2-mercaptoethylamine) to form a highly fluorescent isoindole which is detected by a fluorescence detector.

Holding Time:   28 days from collection to analysis to analysis if samples are properly preserved.
Preservatives:   Sample should not exceed 10°C after the first 48 hours after collection. Store samples at 6°C. Protect from sample from light. Add potassium dihydrogen citrate to yield a concentration in the sample of 9.2 to 9.5 g/L. Add sodium thiosulfate to yield a sample concentration in the range of 80 to 320 mg/L.
Required Preps:   40 or 60mL Amber glass bottle with Teflon-lined lid
